The aim of the present study was to investigate comparatively the in vitro action of recombinant material of TNF-a (rTNF-a) and IFN-y against growth of purified myeloid progenitor cells from normal donors and patients with acute (AML) and stable-phase chronic myelogenous leukemia (CML). Material and Methods Cell Preparation Normal marrow was aspirated from consenting volunteers. Mononuclear cells were isolated by Ficoll gradients. Progenitor cells were enriched by using an immune rosette technique previously described [16].

As shown, two of the seven patients revealed IC50 CFU-GM values much lower than normal controls. One complete and one partial clinical response were achieved and cytogenetic improvement with significant reduction of the frequency of the Ph1_ Effect of Recombinant Interferon-Alpha-2 on the Growth 39 Table 1. Response of PhI-Positive CML in relation to pretreatment IC50 (CFU-GM) Patient No. 0 7 10000 180 600 Response" Cytogenetic improvement +b F(BC) " According to the criteria of Talpaz et al. [9].

In all these patients, additional Pvull fragments could be found with our probe. We do not yet know whether this group of patients, defined on their ber restriction pattern, really represents a distinct subgroup of CML patients. More research has to be done to answer this question. Recently, a correlation between clinical variations and genomic alterations in Ph I_ negative patients has been demonstrated [8]. In contrast to these results, we did not find any significant correlation in our group of mainly PhI_positive patients.
